HOXA10-AS boosts EC cell growth via regulating HOXA10.
(a) RT-qPCR and western blot assays were carried out to detect the expression of HOXA10 in HEEC and EC cell lines. (b) The RNA and protein levels of HOXA10 after HOXA10-AS reduction in EC cells were tested through RT-qPCR and western blot assays. (c) The overexpression efficiency of pcDNA3.1-HOXA10 was determined via RT-qPCR. EC cells were divided into four groups and transfected with different plasmids: sh-NC, sh1-HOXA10-AS, sh1-HOXA10-AS+pcDNA3.1 and sh1-HOXA10-AS+pcDNA3.1-HOXA10. (d-e) Colony formation and CCK-8 assays were performed to evaluate the proliferation and viability of EC cells under different conditions. (f) TUNEL assay was conducted to evaluate the apoptosis of EC cells with different treatments. (g-h) Transwell assays were performed to examine the migratory and invasive abilities of EC cells transfected with different plasmids. **P < 0.01, ***P < 0.001.
