CHX treatment showed negligible effects on JP8BdelER expression and target transcription. Lysates of BL2-ΔC-AIDER cells treated by OHT for 6 h with or without CHX addition 1 h in advance were analyzed by Western blot (A). The intensity of JP8BdelER was measured and normalized according to that of tubulin. Intensity relative to the OHT sample after normalization is shown by percentile. IgH gene transcription was checked by RT-PCR (B) and ChIP with anti-RNA polymerase II (C). The numbers on the RT-PCR gel image indicate the average intensity of each band that is obtained by photoshop (Adobe Systems, San Jose, CA) after image scanning.
