Primer extension analysis of the ysnD transcript. RNA samples prepared from cells of strains FU459 (tnrA::cat) and 168, which had been grown in MM medium containing 13.6 mM sodium glutamate instead of glutamine to an optical density at 600 nm of 0.6, were subjected to primer extension analysis, as described in the text. Lanes 1 and 2 contained the cDNAs extended from the primer by using 50 μg of total RNA from strains FU459 and 168, respectively. Lanes G, A, T, and C contained the products of the corresponding dideoxy sequencing reactions, as described in the text.
