Treatment of fibroblasts from bleomycin-injured mice with HDAC inhibitors resulted in increased expression of Fas. Fibroblasts from saline- and bleomycin-treated mice were treated for 72 h with the DNA methylation inhibitor 5-aza-dC (1 μM) or the HDAC inhibitors TSA (0.1 μM unless otherwise specified) and SAHA (1 μM) and (a) assayed for Fas mRNA levels by real-time RT-PCR (n=4 for saline, n=6 for bleomycin; normalized to untreated control) and (b) cell surface expression by flow cytometry (n=2 for saline, n=4 for bleomycin).
