Effects of NSAIDs on [Ca2+]i. Fura-2-based ratiometric Ca2+ imaging experiments were performed on C6 cells. Various test agents were applied, as indicated by the horizontal bars. Line graphs were obtained by averaging data points from all recorded cells in at least three independent preparations obtained from three different cell passages. A: IBU, even at high concentrations, had no effect on [Ca2+]i. B, C: CELE and SS caused concentration-dependent elevation of [Ca2+]i. The effect of both agents was readily reversible, because the substance-induced [Ca2+]i elevation subsided upon removing the compounds from the bathing solution. D: THAP, 2 μM, shown for comparison, causes the irreversible blockade of SERCA and a [Ca2+]i elevation. CELE, celecoxib; SS, sulindac sulfide; THAP, thapsigargin; IBU, ibuprofen.
