Figure 6. Homology Model for BASS and superimposition onto the EM data.

Figure 6

(a) Homology model of BASS. Black arrows point to the N-terminal end of the parallel dimers. Rosetta modelling was used to construct the model and EPR data were used to refine it. The spins are shown on residue 671 and 791 (indicated by arrows) for the orientation of the chains. According to the model parallel dimers form a tetramer in an antiparallel fashion. The dimer region extending out from tetrameric core starts around residue 835. Each strand of BASS domain is shown with a different colour (b). The homology model; all strands are shown in white and the CC regions supported by EPR analysis are highlighted in green. The regions are between residues 657–671, 708–719 and 776–788. (c) STEM image of full-length Kinesin-5 (left) and BASS (right) decorated with 2 nm Nanogold particles. (d) Superimposition of the homology model on the stem images shown in (c) by taking into account the dimensions of the model. The 2 nm Nanogold decoration at the N-terminus of the BASS matches with the N-terminal location in the homology model.
