Positive feedback relationship between HIF-1 and JMJD2C promotes breast cancer progression. (A) HIF-1 activates transcription of the KDM4C gene encoding JMJD2C, which is recruited by HIF-1α to the HRE of HIF-1 target genes and demethylates H3K9me3, thereby stimulating transactivation of target genes encoding proteins required for glycolytic metabolism and lung metastasis. (B) Knockdown of JMJD2A expression in human breast cancer cells impairs metastasis to the lungs after injection into the mammary fat pad of immunodeficient mice.
