PEGylation reduces cytokine production after vector administration. IL-6 (A), TNF-α (B) and IL-12 p70 (C) were measured in serum samples obtained from animals 6 h after administration of 5 × 1011 or 3 × 1012 vp/kg of either unmodified or PEGylated helper-dependent adenovirus in the non-human primate. Clinical parameters such as platelet counts (D), activated partial thromboplastin time (aPTT) (E), D-dimer (F), white blood cell (WBC) counts (F), granulocytes (G) and lymphocytes (H) were measured over time in baboons given either modified or PEGylated helper-dependent adenovirus expressing beta-galactosidase. Data represent values obtained from one primate per experimental condition.
