Figure 2.
The purinergic receptor subtype designated P2Y2 is involved in propagation of this ICW. The P2Y2 receptor subtype is activated by ATP and UTP and linked to phospholipase C (PLC) activity. A. Typical Fura-2 fluorescence ratio (ΔFratio) traces (left) show the dose dependence of the Ca2+ transients induced by 1 μM to 100 μM ATP (top) and UTP (bottom). Right side shows the corresponding dependence of the peak amplitude of the Fura-2 fluorescence ratio (ΔFratio) as a function of ATP and UTP concentration as indicated. The EC50 values were 1.30±0.05 μM ATP and 0.91±0.06 μM UTP. EC50 values were obtained from the best fit using Hill’s equation, y=Vmax*xn/(kn+xn), where Vmax is maximal response, x is the concentration of agonist, k is EC50, and n is Hill’s coefficient. B. Fura-2 fluorescence ratio (Fratio 340/380) images illustrate the effects on ICW induced by mechanical stimulation of P2Y2 desensitization with 500 μM UTP, inhibition of PLC by U73122, and negative control with its inactive analog U73343. Other conditions as in Figure 1.
