ACTN4 drives multinucleation in NR6WT fibroblasts which is limited by phosphomimetic ACTN4. NR6WT fibroblasts were transfected with WT ACTN4-eGFP or Y4E/Y31E-eGFP followed by 48-h culture with complete growth media. After trypsinization, cells were reseeded on 6-well plates coated with 1 μg/ml fibronectin and then cultured for an additional 2 h prior to fixation. Percentages of dual/multinucleated cells were analyzed (n>200). p < 0.01 comparing cells expressing the WT ACTN4 with those expressing the dual phosphomimetic ACTN4. Error bars indicate ±S.D.
