Induction of DNA fragmentation in cyclooxygenase-null cells (COX-1^−/−/COX-2^−/−) by NSAID treatment. Postconfluent cells were treated with different NSAIDs for 72 h, and genomic DNA was purified and resolved on the 1.5% agarose gel as described in Materials and Methods. Lane 1, 1-kb DNA ladder; lane 2, control postconfluent cells with no treatment of NSAIDs; lanes 3–7, the same cells treated, respectively, with sulindac (200 μM), NS-398 (100 μM), ibuprofen (1 mM), piroxicam (200 μM), or indomethacin (200 μM).
