Dileucine-based motifs bind selectively to the γ/σ1 and the α/σ2 hemicomplex. (A–F) The different subunits of AP-1 and AP-2 were tested for their ability to bind to a panel of dileucine-based motifs derived from the indicated proteins. GST-clathrin TD and GST-YSKV served as positive controls in the binding assays for the β and μ subunits, respectively. Binding assays were performed as described in Materials and Methods, and Western blots were probed with the indicated antibodies. μ1* indicates a 5X longer exposure of the μ1 blot.
