Figure 7.
Status of histone acetylation at the hTERT promoter in ESC and differentiated cells. (A) ChIP analysis of the transgenic promoters in F2-9 ESCs, F2-9d cells, and F2-9d cells treated with 5 μM TSA for 24 h. Antibodies used in ChIP are as follows: 1, acetylated histone H3; 2, acetylated H4; 3, RNA polymerase II; 4, no antibody. Lanes 5-7 are 1/10, 1/20, and 1/40 of input chromatin fragments, respectively. Lane -, no template control. Precipitated DNA fragments were PCR-amplified using primers specific to the hTERT and hCRR9 promoters and analyzed on agarose gels. (B) TSA treatment on luciferase expression in differentiated cells. F2-1d and F2-9d cells were treated with 0, 2.5, or 5.0 μM TSA for 12 or 24 h. The data are average ratios of Fluc to Rluc activities in triplicate wells of 24-well plates. *p < 0.00003.
