Bacteriophage linked assays for antibodies to protein antigens

Abstract

Bacteriophage has been coupled to the monovalent antibody fragment Fab' made by enzymatic digestion of antiphage antibody. Concomitant neutralization of the phage (T4) was slight under the conditions used. The phage could then be neutralized by anti-Fab sera. This method has been used to assay for anti-Fab antibodies and found to have a threshold sensitivity of about 6 ng antibody/ml. It is hoped to couple other protein antigens to phage by prior linkage to anti-phage Fab′.

A method for the rapid titration of large numbers of anti-phage and anti-Fab sera using a phage applicator is described.
