FIG. 1.
(A) Phylogenetic tree and schematic alignment showing the relationship between putative MYST family acetyltransferases, S. pombe (Sp) Mst1p (GenBank accession number AL034583) and Mst2p (GenBank accession number Z69795), human (h) Hbo1p, Tip60p, Mozp, and Morfp, and S. cerevisiae (Sc) Esa1p, Sas2p, and Sas3p. Zinc fingers (zf), chromodomains (chromo), and H1 and PHD domains are indicated. Percentage of identity, similarity, and gaps with respect to the MYST domain of SpMst2p are shown below the MYST domains of each protein (identity/similarity/gap). Numbers above the Mst2p MYST domain indicate amino acids (aa). (B) SpMst1p is essential for cell survival. One copy of the mst1+ gene was disrupted in diploid cells with ura4+. Ura+ transformants were induced to undergo meiosis, and spores were analyzed by tetrad dissection on YES plates. After 3 to 5 days at 25°C, colonies were replica plated to minus uracil plates (−ura) and incubated for 3 to 5 days at room temperature.
