Correct and incorrect 5′-end maturation of precursor tRNAs (cleavage precision) in E. coli BW bacteria depending on protein-only RNase P enzymes (HARPs) for cell growth, analyzed by RNA-seq. The fraction of tRNAs with 5′-ends derived from correct processing by RNase P is plotted for individual tRNA species; tRNA reads starting at the 5′-end position +1 (at position −1 for tRNAHis and tRNASec) were classified as “canonically cleaved,” and reads beginning at −1 (at +1 for tRNAHis and tRNASec) were classified as “miscleaved.” RNA was prepared from cells harvested in exponential growth phase at an OD600 of ∼0.5–0.75 in arabinose-free LB medium and analyzed by RNA-seq (two biological replicates for each complementation strain). Only tRNAs with a total number of reads of at least 100 cpm (counts per million) across all eight libraries (sum; corresponding to two replicates each of libraries Eco rnpB, Aae harp, Mth harp, and Tin harp) were considered.
